Lion’s Mane Mushroom: The Nerve Growth Factor Story
Lion’s Mane has one of the more scientifically interesting mechanisms in this category — a plausible route to stimulating nerve growth factor — paired with one of the smaller human trial bases. Both things are true at once, and worth holding together.

What Lion’s Mane is
Hericium erinaceus is a distinctive white, shaggy mushroom that grows on hardwood trees across North America, Europe and Asia, named for its resemblance to a lion’s mane. It has a long history as a culinary mushroom in Chinese and Japanese cooking, and a separate history in traditional medicine, where it was used for digestive complaints.
Supplements typically use either the fruiting body, the mycelium (the root-like network the mushroom grows from), or both, and this distinction matters — the two contain different concentrations of the compounds researchers are actually interested in.
As a culinary mushroom it has a mild, slightly seafood-like flavour when cooked fresh, sometimes compared to crab or lobster — worth knowing because supplement marketing sometimes implies it is an exotic or rare find, when it is a mushroom sold in Asian grocery stores and increasingly in North American ones too.
The nerve growth factor mechanism
Lion’s Mane contains two families of compounds studied specifically for neurological activity: hericenones, found mainly in the fruiting body, and erinacines, found mainly in the mycelium. Laboratory research has shown both can stimulate the production of nerve growth factor (NGF) — a protein essential for the growth, maintenance and survival of certain neurons.
This is a genuinely interesting mechanism, because NGF decline is implicated in some age-related cognitive changes. But laboratory and animal evidence for a mechanism is not the same as a proven human cognitive benefit, and that gap is real here.
The human trial base is smaller than for Bacopa or Ginkgo. The most-cited study, a Japanese trial in older adults with mild cognitive impairment, found improved scores on a cognitive function scale over sixteen weeks that reversed after supplementation stopped — an interesting result, but from one small trial that needs larger, independent replication before it can be called established.
Safety and interactions
What to check before taking it
- Mushroom or mould allergy. As with any fungal supplement, allergic reaction is possible in people sensitive to mushrooms.
- Diabetes medication. Some laboratory research has examined blood-sugar effects; the human evidence is limited but worth mentioning to your care team if you monitor glucose closely.
- Anticoagulant medication. A small number of studies have examined effects on platelet aggregation; the effect size in humans is not well established, but it is a reasonable thing to flag to a pharmacist.
Reported side effects in the trial literature are minimal, and Lion’s Mane has a long history of food use with no established toxicity at culinary or typical supplement doses. The research is searchable on PubMed.
